Selection of suitable housekeeping genes for expression analysis in glioblastoma using quantitative RT-PCR

2009

Abstract Background Considering the broad variation in the expression of housekeeping genes among tissues and experimental situations, studies using quantitative RT-PCR require strict definition of adequate endogenous controls. For glioblastoma, the most common type of tumor in the central nervous system, there was no previous report regarding this issue. Results Here we show that amongst seven frequently used housekeeping genes TBP and HPRT1 are adequate references for glioblastoma gene expression analysis. Evaluation of the expression levels of 12 target genes utilizing different endogenous controls revealed that the normalization method applied might introduce errors in the estimation of…

Software for weather databases management and construction of reference years

2011

The purpose of this paper is to illustrate a procedure that permits, starting from a sufficiently long database of time series, the construction of a Reference Year (RY) of hourly weather data according to the rules of ISO 15927-4 standard. In order to facilitate the management of the weather database and to allow the users to easily generate the file, an algorithm has been implemented in Microsoft Visual Basic for Application (VBA). By this way, the application of the ISO 15927-4 is possible even using a popular software tool such as Microsoft Excel or Access without any other expensive specialized software. Such tool allows to fulfil all the procedures mentioned in ISO 15927- 4 giving as …

Getting new bronchodilator compounds from molecular topology.

2003

Abstract Molecular topology has been used to select new lead bronchodilator compounds. The main advantage of this method, as compared to others frequently used, is that it does not require a previous explicit knowledge of the mechanism of action (MOA) of the compounds analyzed. A large database (12,000 chemicals) has been examined in this study to find less than 5% compounds with bronchodilator activity. After removing those compounds already described as bronchodilators, we present here the results for 20 among these compounds, some of them showing other pharmacological activities. Some of the compounds selected in this study showed higher relaxation and higher potency than theophylline, w…

A practical framework for data management processes and their evaluation in population-based medical registries.

2013

We present a framework for data management processes in population-based medical registries. Existing guidelines lack the concreteness we deem necessary for them to be of practical use, especially concerning the establishment of new registries. Therefore, we propose adjustments and concretisations with regard to data quality, data privacy, data security and registry purposes.First, we separately elaborate on the issues to be included into the framework and present proposals for their improvements. Thereafter, we provide a framework for medical registries based on quasi-standard-operation procedures. The main result is a concise and scientifically based framework that tries to be both broad a…

Target vs non-target analysis to determine pesticide residues in fruits from Saudi Arabia and influence in potential risk associated with exposure

2017

The occurrence of pesticide residues in fruits was determined by a target method for 62 analytes and a wide scope screening method against a database of 500 pesticides. Limits of quantification (LOQs) were from 0.5 to 6.3 μg kg−1 for the target method and from 0.5 to 119 μg kg−1 for the non-target. Thirty samples: dates, apples, oranges, tangerines, lemons and grapefruits were selected because their high consumption, except lemons and grapefruits that were to cover all citrus fruits. Using the target method, 15 compounds (mostly insecticides and fungicides) out of 62 pesticides (organophosphorus, carbamates, pyrethroids, chloroacetanilides, triazines, triazoles, imidazoles, etc.) were detec…

Comparison between sodium dodecylsulphate and cetyltrimethylammonium bromide as mobile phases in the micellar liquid chromatography determination of …

2004

The retention behaviour of non-steroidal anti-inflammatory drugs (NSAIDs) using micellar mobile phases of sodium dodecylsulphate (SDS) is studied and compared with that observed with micellar mobile phases of cetyltrimethylammonium bromide (CTAB). A liquid chromatographic procedure for the determination of acemetacin, diclofenac, indomethacin, ketoprofen, naproxen and tolmetin in pharmaceutical preparations is described. The proposed system uses a Kromasil C18 analytical column and a solution of 0.15 M SDS at pH 3 with 10% 1-propanol as mobile phase. Under these conditions, the studied NSAIDs elute between 6 and 10 min at a 1 mL min(-1) flow rate. Limits of detection (LOD) are lower than 0.…

Autoantibody detection using indirect immunofluorescence on HEp-2 cells

2009

The detection of autoantibodies is an important element in the diagnosis and monitoring of disease progression in patients with autoimmune diseases. In laboratory diagnostic tests for connective tissue and autoimmune liver diseases, indirect immunofluorescence on HEp-2 cells plays a central role in a multistage diagnostic process. Despite the high quality of diagnostics, findings at different laboratories can differ considerably due to a lack of standardization, as well as subjective factors. The present paper formulates recommendations for the standardized processing and interpretation of the HEp-2 cell test for the detection of non-organ-specific (especially antinuclear) antibodies. It pr…

Can Soft Models Describe Polymer Knots?

2020

Similar to macroscopic ropes and cables, long polymers create knots. We address the fundamental question whether and under which conditions it is possible to describe these intriguing objects with crude models that capture only mesoscale polymer properties. We focus on melts of long polymers which we describe by a model typical for mesoscopic simulations. A worm-like chain model defines the polymer architecture. To describe nonbonded interactions, we deliberately choose a generic "soft" repulsive potential that leads to strongly overlapping monomers and coarse local liquid structure. The soft model is parametrized to accurately reproduce mesoscopic structure and conformations of reference p…

EU dissemination of the provisional ultra-low-temperature scale, PLTS-2000

2003

Following the introduction of the provisional low-temperature scale from 0.9 mK to 1K, PLTS-2000, there is a need for primary and secondary thermometers and fixed points, which can disseminate the scale to users. This paper reports on the progress, within the EU collaborative project ‘ULT Dissemination’, in the development and evaluation of several devices with associated instrumentation. Principal among them are a current-sensing noise thermometer, a CMN thermometer adapted for industrial use, a Coulomb blockade thermometer, a second-sound thermometer, a 3He melting pressure thermometer for a direct realisation of the PLTS-2000. A superconductive reference device has also been developed, a…
